The Battle of Britain (1940) is considered Germany's first major defeat in World War II. While Germany failed to achieve air superiority over Britain and had to postpone Operation Sea Lion (invasion of Britain), it was a significant psychological and strategic setback. The Battle of Stalingrad (1942-43) was later and more decisive, but Britain was first.
